What is AICAR Ribonucleotide?

AICAR ribonucleotide (aminoimidazole carboxamide ribonucleotide or AICAR) is an intermediary product of inosine monophospho synthesis that acts as an agonist for AMP-activated protein kinase. AICA ribonucleotide increase p38 mitogen-activated protein kinase activity in skeletal muscles, stimulates glucose intake and suppress apoptosis by reducing intracellular reactive oxygen species (ROS) production. In experimental studies, administration of AICAR significantly improved endurance-type exercise performance by converting fast-twitch muscle fibers to slow-twitch type, energy-efficient and fat-burning muscle fibers. In addition with GW1516, AICAR exercise-related benefits were observed in laboratory animals without exercising. AICAR studies have reported increased fat loss and endurance in study subjects. AICAR promotes the utilization of nutrient stores for energy biosynthesis. 


Studies reveal that AICAR can reduce inflammation through the increase in the amount of nitrous oxide from the endothelial cells. AICAR-dependent precondition is mediated by ATP-sensitive K+ channel and hemeoxygensae mechanism. The mechanism of action is through the increase of ATP-sensitive K+ channels causing action potential and reducing the levels of calcium ions in the system.AICAR is also known as ZMP, Acadesine and 5-Aminoimidazole-4-carboxamide ribonucleotide. AICAR enter cardiac cells and inhibit production of two enzymes such as adenosine deaminase (ADA) and adenosine kinase (ADK). The latter initiates the transfer of gamma-phosphate from ATP to adenosine that regulates intracellular and extracellular aggregations. AICAR blocks these two enzymes that results in stimulation of glucose intake and increase in circulation. The biochemical changes aid the cell to produce increased energy to fuel the cells.
